It's possible, any extraneous vibrations steals energy from the strings.

The only time it's ever necessary is on Fender style plank pegheads, because even with string trees, frequently there's not enough pressure on the nut to keep it in place like there is on a raked peghead. You take a Les Paul, you'd need a hammer to move the nut once the strings are on it. But on a a plank peghead, I just tack it in place with three tiny dots of CA (super glue). This is completely unnecessary on any guitar with a raked peghead. "
I'd use three drops of super glue and see if it sounds better.
